Abstract

To our knowledge, few studies on the use of sildenafil in humans have been published to date, either in human or animal models. Our current investigation of the effects of sildenafil on male rats has yielded limited results in animal models. A randomized, double-blind, placebo-controlled crossover study was conducted to assess the effects of sildenafil on the locomotor activity of male rats. Male rats, n = 10 were administered a single dose of sildenafil (1 mg/kg) or vehicle (0.9% NaCl) at 12-hr intervals for 4 weeks. An additional set of eight male rats received vehicle for the same period. The incidence of locomotor activity, the body weight, the body water consumption, and the number of days spent in the home cage were assessed using the food schedule. The incidence of the locomotor activity and the body weight were significantly lower in sildenafil-treated rats compared to vehicle-treated rats, but not in n = 8 rats.

Introduction

The aging process is known to result in an increase in the number of male sexual characteristics, such as erectile dysfunction [], and a decrease in female sexual characteristics, such as reproductive problems []. These characteristics of aging may contribute to the development of erectile dysfunction and the risk of sexual disorders such as premature ejaculation []. Sildenafil, which is the active ingredient in Viagra, is a selective inhibitor of phosphodiesterase type 5 (PDE5), an enzyme that is involved in the degradation of cyclic guanosine monophosphate (cGMP) []. Sildenafil has been studied in humans for the first time, but its effects on male rats have not been fully reported. A randomized, double-blind, placebo-controlled crossover study was conducted to assess the effects of sildenafil on male rats. Male rats, n = 10 were administered a single dose of sildenafil (1 mg/kg) or vehicle (0.9% NaCl) at 12-hr intervals.
